Agent 4 initially went to Inkopolis for an unknown reason but got swept up in Marie's quest to save the Great Zapfish and her cousin Callie. In Octo Canyon, Agent 4 completes various missions to retrieve stolen Zapfish, defeating many Octarians along the way. Eventually, Agent 4 confronts DJ Octavio and a brainwashed Callie, rescuing the Great Zapfish, freeing Callie, and finally restoring Inkopolis' power supply.

Marie returns as a major player in Splatoon 2's hero mode in Octo Canyon. This time, she is more open about her identity as Agent 2 and fills Cap'n Cuttlefish's role as Agent 4's main advisor and guide, as the Cap'n is out on a research trip with Agent 3. As Agent 4 fights through the Octarian complex in search of the Great Zapfish, Marie reveals to the player that Callie went missing at the same time as the Great Zapfish, and suspects Octarian involvement. After beating the The Octo Shower, she and Agent 4 receive a call telling them to leave Octo Canyon and never look back. However, Marie quickly realizes that the caller is Callie due to the fact that the caller is holding the phone upside down (a recurring gag in the original Splatoon single player). In the final boss battle, it is revealed to the player and Marie that Callie has been brainwashed by DJ Octavio using his hypnoshades and are forced to fight him and Callie; as the battle continues, Marie is able to shoot off the hypnoshades with a Hero Charger filled with "Low-Tide Ink", slowly bringing Callie back to her senses. When Callie finally makes a full recovery, Marie is able to pull out a boom box playing Calamari Inkantation, allowing them to distract DJ Octavio and letting Agent 4 to finish him off, saving the Great Zapfish.

In Splatoon 2 Callie's disappearance at the same time as the Great Zapfish's alerts Marie to the return of DJ Octavio, leading to the recruitment of Agent 4. As the new agent makes their way through Octo Canyon, Marie begins receiving peculiar radio messages that are later revealed to be coming from Callie. When Agent 4 and Marie finally discover Callie, they learn that she has fallen under the influence of DJ Octavio's "hypnoshades" and is now working as one of his subordinates. During the following battle, Marie manages to shoot off the hypnoshades with her Hero Charger, bringing Callie back to her senses.

In Splatoon 2, he once again the final boss, fought in the level Tide Goes Out. the DJ's flying machine has received an upgrade since the last game. It now has a second level, on which a brainwashed Callie stands and sings. Below her is the main control, in which the DJ sits. In front of the turntable and wasabi stalks is a tray of calamari bombs, which go flying (Aesthetically) as the player attacks the DJ. On the back of the machine are some exhaust pipes, which Octavio uses for the dash punch attack. The two giant fists are now gold and sparkly, and have brass knuckles on them, which reads something in the Octarian language. There are 4 holes on the front of the machine, in which bombs and the ink shower attack are deployed from. There are plenty of speakers on the machine, and a plethora of lights as well. The DJ's appearance has also changed: His kabuto helmet is now black with golden accents, and he wears a pair of shutter shades.

Pearl speaks rather bluntly and uses various slang, in line with her characterization as a rapper (despite her "pop princess" appearance). Compared to her partner, Marina, Pearl is a lot more expressive, quick to jump onto a topic with a bit of a short temper, and has a knack for composing new songs as they come to her. Often times, her comments can be playfully sarcastic, playing off her co-host. Her lively attitude is most obvious if the player looks at her through the window, as she looks at them quizzically before striking a pose. Whenever she wins a Splatfest, she boasts how her team was better in the form of a rap, essentially rubbing it Marina's face.

Marina is an Octoling, the Octarian version of an Inkling. She has dark/brown skin, silverish teal eyes and reddish-pink pupils in the shape of an infinity symbol, similar to octopuses in real life, two eyelashes on each eye, downturned black eyebrows and a beauty mark at the left of her lips. She appears to be taller than most Octolings and Inklings. Like Pearl, and unlike other Inklings, she has one sharp tooth in the middle at the top and bottom of her teeth instead of the usual two on top. Her tentacles are purplish brown with teal edges, which are long and worn all the way down reaching past her waist, and her fingers are pointed and teal at the tips, giving somewhat the impression of painted nails. Her outfit consists of a sleeveless black top covering her chest with a large, mostly undone silver zipper with an “8”-shaped hole and a high collar, leaving her midriff visible, showing a teal navel piercing. She has black shorts with an asymmetrical/diagonal waistline and slanted deep teal lines over teal tights, black fingerless gloves, and wears black boots with big undone silver zippers with “8”-shaped holes, similar to the one on her top. She appears to have on large, black headphones with teal rings on the sides of them. Her teal color will change depending on the Splatfest team she chooses, with the exception of her eyebrows.
