Light Yagami is the main protagonist of the Death Note series. After discovering the Death Note, he decides to use it to rid the world of criminals. His killings are eventually labelled by the people living in Japan as the work of "Kira."

Light is characterized as hardworking, talented, and a natural genius. Highly perceptive and good with problem-solving skills, Light is a skilled planner and good at mapping out scenarios. Furthermore, Light is very popular among his peers and adored by his family members. However, his high intelligence, academic achievements, and constant praise from adults laid the foundation for an innate hubris and arrogance that quickly escalated after he adopted the role of Kira. Light also struggled with a feeling of boredom that was quelled once he began using the notebook.

 L is a world-renowned detective who takes on the challenge of catching the mass murderer known as Kira. In his investigation, L becomes suspicious of Light Yagami and makes it his goal to prove that Light is Kira.

L is quite secretive, and only communicates with the world through his assistant, Watari. He never shows his face to the world in person, instead representing himself with a capital letter L drawn in "Old English MT" or "Cloister Black" typeface. It is more likely to be "Cloister Black" as Watari's "W" is different in "Old English MT."

Rem is the Shinigami who gives Misa Amane one of the Death Notes she had. Like Ryuk, Rem possesses two Death Notes; however, Rem did not get hers through trickery. The Shinigami Gelus, who had fallen in love with Misa, sacrificed himself to kill her destined murderer. In doing so he was reduced to dust, leaving only his Death Note. Rem delivered this Death Note to Misa, since it was her life he saved.

While Ryuk takes amusement in everything in the Human World, Rem is almost the exact opposite, viewing most humans with contempt. Also, while Ryuk is ambivalent to Light's success or failure, Rem actively assists Misa. This makes her equally as guilty as Ryuk of not displaying 'proper' traits for a Shinigami to have. She is even willing to sacrifice her life to defend Misa, as evidenced by her threat to kill Light should Misa die before her time. For Misa's sake, however, she still assists Light in his schemes.

Mello is the older of L's two successors raised at Wammy's House, Watari's orphanage for gifted children in Winchester, England. When Roger asks that Mello and Near work together to capture Kira, Mello refuses, pointing out the differences between them. He leaves the orphanage soon after and eventually seeks help from the Mafia.

While Mello is certainly a genius and more intelligent than most people, the concept of being only the second most intelligent person in Wammy's House, behind Near, fuels the inferiority complex that defines Mello's character. Mello verges on immorality in his obsession with being the one to take down Kira and is willing to do "whatever it takes," including kidnapping Sayu Yagami, the Japanese Police Director's daughter, and orchestrating the murders of most of the SPK members. In the manga, after Mello has successfully stolen a Death Note, he even goes so far as to attempt to blackmail the president of the United States into giving him funds and resources to aid his pursuit of Kira and to evade Near, threatening that if his demands are not met he will use the Death Note to force the president to launch the United States' nuclear missiles and start World War III. Arguably, Mello is not trying to defeat Kira out of a sense of "justice" but because capturing Kira will prove to the world that Mello is indeed the worthy successor to L as the world's greatest detective and not simply a runner-up to Near. As Mello describes his goal in his own words, "I will become number one!"

Near is the younger of L's two successors, raised in Wammy's House—Watari's orphanage for gifted children in Winchester, England. After L's death, Near begins his investigation of the Kira case, gathering evidence over a period of four years. Near takes his findings to the President of the United States and reveals himself to be the true successor of L. Soon after, Near becomes the head of the Special Provision for Kira and continues the hunt for Kira. After making contact with "L" (Light Yagami), Near begins to suspect that he is also Kira.

Near is very intelligent, although his childish appearance and demeanor can cause others, such as David Hoope, to question his abilities. He has an obsession with solving complex problems and views an investigation as solving a puzzle. He is highly observational and was once seen piecing together a puzzle that was almost entirely colored white. He is proficient when it comes to multitasking and is able to monitor multiple computer screens at once. Near also appears to be ambidextrous, playing darts with his left hand and stacking dice with his right.

Teru Mikami is a criminal prosecutor chosen as the Hand of Kira by Light Yagami. Mikami takes ownership of Gelus's Death Note after Light falls under suspicion of being Kira again, and needs someone to act as Kira without being given every order.

Mikami is characterized as having a strong sense of justice that has developed over the course of his childhood. He detests crime and, similar to Light, expresses a fervent passion for punishing evil. His sense of justice, however, has developed into a rather "black and white" understanding of people, and he goes as far as to label everyone he meets as either good or evil. Mikami is also notable for his devotion as a Kira-worshipper; his answer to a Kira’s Kingdom survey question that asked what he thought of Kira was simply "God." His loyalty to Kira is exemplified when he forces himself to stop questioning Kira after he is perplexed by the task of creating and using a fake notebook.

Watari is L's handler and the supplier of logistics to the Japanese Task Force. He is also the founder of Wammy's House, an orphanage that has produced some of the best detective minds in the world, including L, Beyond Birthday, Mello, Near, and Matt.

Watari is a famous inventor and the founder of Wammy's House, an orphanage for gifted children in Winchester, England. Using the fortune he made as an inventor, he built an orphanage that teaches children with special talents and sends them out into the world. Before the Task Force is formed, he is the only person known to have seen L and the only one capable of contacting L directly. Like L, he represents himself with a Cloister Black "W" on computer screens. Since L's time at Wammy's House, Watari has been a fatherly figure to him, though in his role as L's handler he acts as a butler. Watari is well-trained in espionage and marksmanship. According to Death Note 13: How to Read, Watari-Senpai likes Earl Grey tea and dislikes dirty rooms.

Naomi Misora is a former FBI agent and the fiancée of agent Raye Penber. After Kira kills Raye and the other twelve FBI agents assisting L with his investigation in Japan, Naomi begins her own investigation.

Described by Death Note 13: How to Read as formerly a "first rate" FBI agent with "remarkable investigative ability," Naomi left the Bureau in October 2003 for the sake of starting a family in the United States. Naomi is very intelligent. Unfortunately, she eventually lets her feelings get in the way of her investigative abilities.

He has a strong sense of duty, and even after the police stop funding the Kira investigation he continues trying to solve the case with the Task Force.

Kiyomi Takada is the former college classmate and girlfriend of Light Yagami who later becomes Kira's spokeswoman on the NHN television station.

Touta Matsuda is a young member of the NPA and the Japanese Task Force.

Matsuda's police skills are lacking in experience, and at times, particularly in the beginning of the series, he somewhat impedes the investigation process. Much to the irritation of his peers, his brash attitude can get him into tricky situations, as evidenced by the time he infiltrated Yotsuba and had to fake his own death to get out. Members of the Task Force, such as Ryuzaki and Shuichi Aizawa, seem to view him as a reckless, naive young detective lacking seriousness for his work.

Sayu Yagami is the kind-hearted younger sister of Light Yagami who plays a minor role in the main storyline, remaining unaware of Light's possession of the Death Note and his role as Kira.

In line with Sayu's age and grade in school she demonstrates age-appropriate behavior, such as watching shows starring Hideki Ryuga and asking Light for help with math homework. She seems to look up to Light, as she recognizes his intelligence and believes that he will become a quality detective. Though they are fairly close at the start of the series, their relationship is somewhat distanced as Light becomes more absorbed into his role as Kira.

Shuichi Aizawa is a member of the NPA and the Japanese Task Force.

Aizawa is an intelligent, respected member of both the NPA and Task Force. He takes his work very seriously and, for this reason, does not approve of Misa Amane's antics. He becomes annoyed when she or someone else refers to her as Misa-Misa, and he often chides Touta Matsuda for his immature behavior. Aizawa's respect for L is grudging, as Aizawa chooses to work within the law and L's methods of investigation are sometimes outside of the law. His contemptuous attitude intensifies after NPA funding for the Task Force is cut, when he conflicts with L over L's withholding of the vital fact that Watari would provide the Task Force with financial support if they chose to stay. He leaves the Task Force for a short while but returns before L's death. After Light Yagami becomes leader of the Task Force, Aizawa is the first member to suspect that Light is Kira. His lawful beliefs are exemplified when he stops Matsuda from killing Light in the final episode.

Gevanni is very talented, having skills which include lock-picking, investigating, monitoring and following suspects, and the ability to forge writing. Even after numerous deaths within the SPK, Gevanni remains a loyal member; this is indicative of his trust in Near. Moreover, he plays a key role in Near's plans and becomes essential to solving the Kira case by following Teru Mikami and subsequently forging a Death Note.

Sachiko Yagami is the wife of Soichiro Yagami and the mother of Light Yagami and Sayu Yagami who is oblivious to Light's ownership and use of the Death Note.

Sachiko is a kind and caring woman who deeply loves her family where she worries about Soichiro Yagami's well-being while he works on the Kira case. When Soichiro is hospitalized due to a stress-related heart attack, she is frequently by his side to care for him. As concerned as she is about Soichiro, she also understands his convictions in working on the Kira case, and she stays by his side. Sachiko is proud of the academic success of her son, Light, and congratulates him on his good grades. When Sayu Yagami is wheelchair-ridden due to emotional trauma caused by her kidnapping, Sachiko aides her recovery, encouraging her and taking her for walks outside.
