Ayano Aishi is the protagonist of Yandere Simulator. She is known as "Yandere-chan" to players of the game, and has been given the cute nickname "Yan-chan" by her classmates.

Since early childhood, Ayano has suffered from an inexplicable, incurable condition. In short, this condition leaves her with a perpetual sense of being empty, hollow, and incomplete. As a result, she has never been able to experience satisfaction, fulfillment, or happiness, and has not been able to experience emotions in the same capacity as other people. As she grew older, she learned to pretend to be a normal person in order to avoid drawing negative attention from others, but was never able to feel any degree of empathy towards other human beings.

Ayano attends a post-highschool academy called "Akademi." On the first day of her second year at Akademi, Ayano came into physical contact with a young man named Taro Yamada. In that moment, she gained the ability to experience everything that had been missing from her life; she felt complete, she felt fulfilled, and she felt powerful emotions. Meeting Taro was like seeing color for the first time, hearing music for the first time, and feeling warmth for the first time. In his presence, she felt overwhelmed with euphoria...but, as soon as they were apart, the feelings of emptiness returned. After just one meeting, Ayano had become hopelessly addicted to Taro's presence, and came to rely on him in order to feel alive.

Then, Ayano saw something horrible...something that terrified her and infuriated her at the same time. It was a young woman who clearly had deep feelings for Taro. If Taro entered a relationship with this girl, then Ayano might lose him forever...

Without a moment's hesitation, Ayano knew exactly what had to be done. This girl - her rival - had to be eliminated as soon as possible, by any means necessary, no matter what the cost.

The Japanese word "senpai" refers to someone who has seniority over someone else. For example, if you are a first-year student, all second-year and third-year students are your "senpai". If you start working at a new job, any co-worker who has been employed at the workplace longer than you is your "senpai".

In Yandere Simulator, the title of "Senpai" is given to the young man (or young woman) that the protagonist is infatuated with. The player is given the option of choosing Senpai's gender at the beginning of the game; they might be "Senpai-kun", or they might be "Senpai-chan". If Senpai is a male, his name is Taro Yamada, and if she is a female, her name is Taeko Yamada.

Over the course of the school year, ten different girls will fall in love with Senpai. There is a myth at school that if a girl confesses her love to a boy under the cherry tree behind the school on a Friday, the two of them will love each other forever. Every girl who falls in love with Senpai plans to confess at 6:00 PM on Friday. If there is anyone who wants to keep Senpai all to herself, she has until 6:00 PM on Friday to eliminate the competition.

Sakyu Basu and her younger half-sister, Inkyu Basu, are a pair of young women who recently enrolled in Akademi. Their scores on the entrance exam were exceptionally high, as if they possessed knowledge far beyond their apparent ages. However, after classes began, their academic performance dropped to an average level, as if they were attempting to avoid drawing attention to themselves.

Sakyu's calm and mature nature contrasts greatly against the energetic and childlike behavior of her younger sister. Although both young women are friendly enough when approached by others, they usually keep to themselves, preferring each other's company above anything else. Despite their friendly demeanor, there are many students at Akademi who feel inexplicably intimidated by them.

When asked why they chose to attend Akademi, they always provide the same answer: "We're looking for something." This woefully short reply is far too vague to adequately satisfy the curiosity of anyone who wants to learn more about them. However, no matter how much they are begged to elaborate, they refuse to say anything more about their intentions at Akademi.

Kizana is the third rival and president of the Drama Club. She's based on the "himedere" archetype, being a snobbish and arrogant person. She dreams of becoming a famous actress and strives to be at the top of every list, be it popularity, school grades, or club funding. Despite being a rather popular girl at school, most people get annoyed at her for her egoistical behavior to be actual friends with her unless they admire her acting abilities enough to ignore her flaws.

Kizana meets Taro in front of a theater when he's feeling at his lowest after losing Amai, and immediately marks him as a perfect candidate for the lead role in her next play. Even though Taro doesn't explicitly agree to participate, Kizana practically forces him, as in her words, he "must and burn his as fuel to elevate his next artistic creation to greater heights". She shows only her good side around Taro, hiding her true nature from him; so despite considering Kizana a bit bossy, Taro will be impressed by her acting abilities and be flustered by what she thinks of him.

Asu is the fifth rival and president of the Sports Club. She's based on "genki" archetype and has a cheerful and energetic personality. She also has a competitive side to her, so she will spend the first four weeks training for Olympics at a specialized institution.

She will meet Taro when he is feeling drained after the first four weeks and will persuade him to do sports together, which appears to be her "love language".

Muja is a student nurse and will appear in Akademi as she will be receiving training from Nasu Kankoshi. Taro will be sick during her week, so he will have to visit her every day. She is a very physically attractive woman, which could help Taro fall for her, but the other factor is that she is very caring and interested in healing and helping others. In addition to falling in love with him, Muja will also develop respect for Taro because he knows a lot of things that Muja herself had to learn in school.

Mida is the seventh rival and a teacher substituting Kaho Kanokogi. Being based on a "sexy teacher" trope, she's portrayed as a villain character, getting a thrill from seeing the boys developing crushes on her even though she knows it is wrong.

Mida will pursue Taro because she is ready to "settle down". Her week will revolve around Mida trying to give him more confidence and courage around girls by roleplaying with him and acting out various scenarios. In these scenarios, she pretends to be a cute younger student (kohai) who looks up to him (senpai). Despite that Taro normally doesn't like women like Mida, and their potential relationship would be very complicated, he won't be able to resist her charms.

Osoro is the eighth rival and leader of the delinquents. To most people, she's known as the strongest and most dangerous girl in school, who had allegedly beaten a number of people on her first day at school. In reality, her closed-off and intimidating persona is most likely the result of her growing up in a poor and abusive family. She had no close friends prior to her current "cronies", and has no interested in making friends in present time; however, she seems to have warmed up to them over time.

Osoro will not be present for the first seven weeks as Genka Kunahito, the guidance counselor, had suspended her in order to be able to reform the delinquents while their biggest negative influence, Osoro herself, isn't present at Akademi. The exact way how she will meet Senpai and what he sees in her is currently unknown, however, Osoro is hinted to have a "different side", most evident in the alternative line in the introductions video and her relationship with her "cronies", as the structure of the group reminds much less of a gang than a club or a friend group.

Megami is the tenth rival and president of the Student Council. She's based on Mary Sue trope, being perfect in almost every possible aspect, from general and academic knowledge to looks and any task that doesn't require years of experience and knowledge.[31] She is a future CEO of Saikou Corp, a multinational corporation that has a monopoly over many industries in Yandere Simulator's world. Her grandfather, Saisho Saikou, held strong beliefs in eugenics and nationalism and controlled everything in his family, including his children's marriage and the way they raise children; this resulted in Megami being just "human" enough to not turn off people like her father, but otherwise superior to almost all other people.

Megami is currently forbidden from attending school during the first nine weeks for unknown reasons. She seems to be aware of Ayano's true nature, and her actions will most likely be directed more towards Ayano than Taro. However, it is currently unknown whether or not she actually has any personal feelings towards Taro.

Kokona Haruka is a cheerful and friendly young woman who participates in the Drama Club at Akademi.

She is known around school for her eagerness to help others in need. If the Cooking Club needs someone to taste-test a new dish, she is the first to raise her hand. If the Science Club needs a test subject for their latest dangerous experiment, she volunteers without hesitation. This has led a few students to give her unflattering nicknames such as "test dummy" and "guinea pig," but she doesn't really see this as anything to be ashamed of.

Kokona joined the Drama Club because of her deep admiration for the club's leader, Kizana Sunobu. (This admiration runs so deep that Kokona is willing to tolerate or outright ignore Kizana's numerous personality flaws.) Kokona's "twin drill" hairstyle was inspired by Kizana, who often styles her hair into very large ringlets.

Kokona enjoys stageplays immensely, and has begun writing a play of her own - a story about a serial killer. Despite the fact that Kokona is a very bubbly and lighthearted person, her serial killer play is actually quite dark and morbid, which surprised her fellow clubmembers a lot. Kizana considers Kokona's play to be distasteful, and has strongly discouraged her from pursuing it.

Kokona is completely unaware that one of the other students at school is actually the daughter of the loan shark that is extorting her father. Despite her friendly nature, Kokona feels such intense anger towards that loan shark that - if manipulated or provoked in the right way - it's entirely possible that she could be convinced to harm or even kill the daughter of that loan shark...
