John Winchester was Samantha and Deanna's father, and son of Henry Winchester.
John and Mary Campbell [The girl's mother] did not initially get along when they met. However Heaven was interested in bringing together the lines of hunters and Men of Letters, and so a Cupid intervened and they fell in love John and Mary's marriage was not always happy, and at least once he moved out of the family home. When Mary died in 1983, John raised their two daughters, Samantha and Deanna alone. Due to the nature of her death, John sought revenge, taking up the occupation and lifestyle of a hunter, which created a unique childhood for his two young children. John moved his family from town to town following leads on supernatural creatures, often leaving the girls for days or even weeks to fend for themselves.
John cut off contact with Samantha after Samantha chose to leave the family and hunting and attend Stanford, although he regularly visited Palo Alto to check up on Samantha. In 2005, John got a lead on the demon that was responsible for Mary's death, and he left Deanna in order to pursue it.
He eventually reunited with Samantha and Deanna in order to obtain the Colt. As they got closer to the Yellow-Eyed Demon, John was kidnapped by the demon Max, and possessed by Azazell. However Samantha wasn't unable to kill her father with the Colt in order to kill the demon.
Following a car crash caused by a demon, John offered his own life and the Colt to Azazell to save Deanna. John revealed that he has known the "truth" about Samantha all along. Before he died, John warned Deanna that she may have to kill Samantha.

JOHN
So... You girls really wanna know about this Colt?
SAMANTHA
Yes sir.
JOHN
It's just a story. A legend, really. Well, I thought it was. Never really believed it until I read Daniel's letter. Back in 1835, when Haley's Comet was overhead, the same night those men died at the Alamo, they say Samuel Colt made a gun. A special gun. He made it for a hunter, a man like us, only on horseback. The story goes, he made thirteen bullets. This hunter, he used the gun a half-dozen times before he disappeared, the gun along with him. 'Til somehow Daniel got his hands on it. They say... they say this gun can kill anything.
DEANNA
Kill anything, like supernatural anything?
SAMANTHA
Like the demon.
JOHN
Yeah, the demon. Ever since I picked up its trail, I been lookin' for a way to destroy that thing. Find the gun, we may have it.
This is John talking to Samantha about her education, and admits why she had the opinions that she did. You can either read Samantha's lines or ignore them.
JOHN
I don't think I ever told you this, but... the day you were born, you know what I did?
SAMANTHA
No.
JOHN
I put a hundred bucks into a savings account for you. I did the same thing for your sister. It was a college fund. And every month I'd put in another hundred dollars. Until... Anyway, my point is, Samantha, that, this is never the life that I wanted for you.
SAMANTHA
Then why'd you get so mad that I left?
JOHN
You gotta understand somethin'. After your mother passed, all I saw was evil. Everywhere. And all I cared about was, was keepin' you girls alive. I wanted you prepared. Ready. So somewhere along the line I uh, I stopped being your father, and I, I became your, your drill-sergeant. So when you said that you wanted to go away to school, all I could think about, my only thought was, that you were gonna be alone. Vulnerable. Sammy it just, it never occurred to me what you wanted. I just couldn't accept the fact that you and me... we're just different.
[Samantha laughs]
JOHN
What?
SAMANTHA
We're not different. Not anymore. With what happened to Mom and Jack... hell we probably have a lot more in common than just about anyone.
JOHN
I guess you're right, girl.
SAMANTHA
Hey Dad... whatever happened to that college fund?
JOHN
Spent it on ammo.
[They laugh]
This is when John is possesed by a demon who is planning on killing Samantha and Deanna. You can either read Deanna's lines or ignore them.
JOHN
Your dad, he's in here with me. Trapped inside his own meat suit. He says "hi" by the way. He's gonna tear you apart. He's gonna taste the iron in your blood.
DEANNA
Let him go, you son of a bitch. Or, I swear to God...
JOHN
What? What are you and God gonna do? You see, as far as I'm concerned, this is justice. You know that little exorcism of yours? That was my boy.
DEANNA
Who? Max?JOHN
The one in the alley? That was my daughter. You understand?
DEANNA
You gotta be kidding me.
JOHN
What? You're the only one that can have a family? You destroyed my children. How would you feel if I killed your family? Oh, that's right. I forgot. I did. Still, two wrongs don't make a right.
