Twilight Sparkle is the central main character of My Little Pony Friendship is Magic. She is a female unicorn pony who transforms into an Alicorn and becomes a princess in Magical Mystery Cure.
"You see, Nightmare Moon, when those Elements are ignited by the... the spark, that resides in the heart of us all, it creates the sixth element: the element of... magic!"
"As the Princess of Friendship, I try to set an example for all of Equestria. But today, it was Spike who taught me that a new friend can come from anywhere. I guess everypony still has things to learn about friendship. Even me! And if Spike says Thorax is his friend, then he's my friend too."

Pinkie Pie, full name Pinkamena Diane Pie, is a female Earth pony and one of the main characters of My Little Pony Friendship is Magic. She is an energetic and sociable baker at Sugarcube Corner, where she lives on the second floor with her toothless pet alligator Gummy, and she represents the element of laughter.
"Exactly! That's why I need you gals to drop what you're doing and help me find some maracas!"
"You see, I saw you when you first got here, remember? You were all, 'Hello,' and I was all, [gasp], remember? You see, I never saw you before, and if I never saw you before that means you're new, 'cause I know everypony, and I mean everypony in Ponyville, and if you're new, it means you haven't met anyone yet, and if you haven't met anyone yet, you must not have any friends, and if you don't have any friends you must be lonely, and that made me so sad, and I had an idea, and that's why I went [gasp], I'll just throw a great big, ginormous, super-duper spectacular welcome party and invite everyone in Ponyville. See? And now you have lots and lots of friends!"

Rainbow Dash is a female Pegasus pony and one of the main characters in My Little Pony Friendship is Magic. She maintains the weather and clears the skies in Ponyville. As a huge fan of the Wonderbolts, she becomes a reservist member of the elite flying group in Testing Testing 1, 2, 3 and a full member in Newbie Dash.
"The Wonderbolts! They're gonna perform at the celebration tomorrow, and I'm gonna show 'em my stuff."
"The shape's fine. Just make the whole thing, you know, cooler. [pause] It needs to be about 20% cooler."
"AAAAAHHH! What do I do? Everypony's gonna see me fail! The Wonderbolts will never let a loser like me join! Princess Celestia will probably banish me to the Everfree Forest! MY LIFE IS RUINED!!"

Princess Celestia, called Queen Celestia in one comic and early development, is an Alicorn pony, the former co-ruler of Equestria alongside her younger sister Princess Luna, and the adoptive aunt of Princess Cadance. In addition to her responsibility of raising the sun, she has also been the teacher of Twilight Sparkle and Sunset Shimmer at different periods in time, often addressing Twilight as "my faithful student" while mentoring her.
"That chamber is protected by a powerful spell that only I can break! This doesn’t make sense."
"I told you that you needed to make some friends — nothing more. I saw the signs of Nightmare Moon’s return and I knew it was you who had the magic inside to defeat her, but you could not unleash it until you let true friendship into your heart."

Apple Bloom is a female school-age Earth pony and a member of the Apple family. She is the younger sister of Applejack and Big McIntosh and granddaughter of Granny Smith. Apple Bloom is one of the founding members of the Cutie Mark Crusaders, a club/"secret" society devoted to helping ponies earn their cutie marks, along with Sweetie Belle and Scootaloo.

Gilda is a female griffon and supporting character in the series. She first appears as an antagonist in Griffon the Brush Off, and she makes subsequent appearances in later episodes,

Zecora is a female zebra who resides in a hut in the Everfree Forest and first appears in Bridle Gossip. Initially believed to be an evil enchantress by the terrified populace of Ponyville, she later befriends the ponies and frequently helps them out with her knowledge of magical ailments.

Spike, also known as Spike the Dragon, is a male "pre-teen" dragon and one of the seven main characters of My Little Pony Friendship is Magic. He is Twilight Sparkle's best friend and number one assistant. He has the ability to magically deliver scrolls to and from Princess Celestia with his fire breath, which features in many episodes, as does his crush on Rarity.
"Well, actually, this is my first birthday in Ponyville. I usually just get one present—from Twilight—a book."
"So...no matter what happens, you're not gonna move a muscle, huh? Then maybe you won't mind if I... eat an entire tub of ice cream!"
