Autumn Leaves is a short romance VN. It follows the main character as he moves to Japan to live with a family trio. Things go awry when he meets his estranged half-sister,
The VN is short, contains three routes-- two male routes and a female. Keep in mind that the two male characters listed are the Yaoi Routes

Itsuki is the eldest of the Akiba siblings, at age 27. He is cold and tsunderes very frequently. He isn't very happy about the main character joining his family, and is even less happy to find out who he is.
His voice should be that of a bishonen character. Note that his path is Yaoi.
[Coldly] I don't accept this. I don't care what Setsuna told you, you are not, nor will you ever be part of my family.
[Angry] It's YOUR fault that we're stuck out here. So what are you going to do, Mr. Hero? If this is how badly you can ruin my day, I can only imagine how Hikari feels. You ruined her damn life! Go back to wherever the hell you came from and leave her alone!
[Sweetly] You've got to be more careful. Jeez, you make even a clutz like Ren look orderly. *pause* All better? Good. Do something stupid like that again and you'll regret it, got it?
Ren is the second child in the Akiba family at age 25. He is a crossdresser who should sound feminine when he speaks while crossdressing. (Think Haruhi's dad from Ouran).
When he is not, he is cheeky and flirtatious. He also uses Japanese honorifics such as '-nii' and '-chan' when referring to his older and younger sibling. He is indeed romancible, making his route a Yaoi route.
If no one is able to pull this off, two different people will be casted for the role.
[Crossdressing] "Oh, honey. Look at how precious you are~ I have a coat that would look devine on you!"
[Himself] "Itsu-nii? Why are you being so mean to him? Maybe he was just trying to make up with Hi-chan? I'm sure Se-chan wouldn't approve at all~!"
[Flirtatiously] "You're absolutely stunning today. Are you dressing up just for me? Or is Itsu-nii the one who's the object of your desire?"
